Summary We are examining the mechanisms by which the nuclear proteins PARP-1 and NFkB (subunit p60) regulate gene expression in human cardiomyocytes in response to stress from cytokines (in this case, TNFalpha or TNFa). We performed ChIP-chip for PARP-1, NFkBp60, RNA Polymerase II (Pol2) and histone H3 in AC16 human cardiomyocytes, and looked at 23550 RefSeq genes to determine localization of these factors. Each experiment was performed in duplicate, except for the H3 ChIP-chips, which were performed once.
Overall Design ChIP-chip biological replicates for PARP-1, NFkBp60 and Pol2 (2 replicates each), and one replicate for H3, from AC16 human ventricular cardiomyocytes cells are included.
